Not in any living Indigenous American population included in the researchers’ comparative dataset.

A previously undocumented branch of the early South American population radiation, sitting in a high-altitude valley in Colombia, preserved in the bones of hunter-gatherers who had lived there thousands of years before anyone thought to look.

And the genetic signature of those people — the specific ancestral fingerprint that distinguished the Checua hunter-gatherers from every other population in the study’s comparison set — showed no detectable contribution to the later ancient populations from the same region, or to any of the present-day South American populations the researchers tested.

The lineage, as far as the current evidence goes, vanished from the genetic record.

Ancient DNA research — the extraction and sequencing of genetic material from prehistoric human remains — has transformed the study of human prehistory over the past fifteen years.

The technology that made this possible was the development of next-generation sequencing: the ability to read millions of short, degraded DNA fragments simultaneously, reassembling them computationally into something that reflects the genetic makeup of the ancient individual who left them behind.

Using these methods, researchers have built an increasingly detailed picture of how the Americas came to be populated.

They have sequenced genomes from early North Americans, including the famous Anzick-1 individual associated with the Clovis culture in Montana, one of the oldest genomes yet recovered from the Western Hemisphere.

They have analyzed individuals from the Pacific Coast of North America, including ancient inhabitants of the California Channel Islands, who represent a distinct ancestral lineage.

They have sequenced genomes from ancient Peruvians, ancient Brazilians, ancient Argentinians, ancient Chileans, ancient Bolivians.

The Americas, as a subject of ancient DNA study, are increasingly well-mapped — but with significant regional gaps.
